Melanotan 1 (MT1) 10 mg

Melanotan 1 (MT-1) is a synthetic linear analog of alpha-melanocyte-stimulating hormone (α-MSH). Developed at the University of Arizona in the 1980s, it substitutes norleucine at position 4 and D-phenylalanine at position 7 of the natural 13-amino-acid sequence, changes that give the analog markedly greater stability than α-MSH itself. The same sequence is known in the literature as [Nle4, D-Phe7]-α-MSH and by the INN afamelanotide.

MT-1 is distinct from Melanotan 2: MT-1 is a linear tridecapeptide, while Melanotan 2 is a shorter cyclic heptapeptide with a different receptor-selectivity profile.

Melanocortin-1 Receptor (MC1R) Agonism

MT-1 is a high-affinity agonist at the melanocortin-1 receptor expressed on melanocytes. Receptor activation is coupled to adenylate cyclase and a rise in intracellular cAMP, which is the signaling readout most often measured in MC1R pharmacology.

Melanogenesis in Cell Culture

In cultured melanocytes and melanoma cell lines, MC1R activation by MT-1 is followed by changes in tyrosinase expression and eumelanin synthesis, the observations that define the compound’s use as a melanogenesis reference agonist.

Receptor Selectivity

Compared with Melanotan 2, MT-1 shows greater selectivity for MC1R over the MC3R and MC4R subtypes, which makes the pair useful together in melanocortin receptor-subtype studies.

MT-1 is used as a reference MC1R agonist in receptor pharmacology, in melanocyte and pigmentation cell-culture models, and in comparative studies of α-MSH analogs.
